چکیده

Abstract Background Clinical and laboratory markers have been exploited to model risk in patients with primary CNS lymphoma (PCNSL), but the derived models do not fully explain observed variation outcome. Here we present an extended framework of phenotype-epigenotype correlations that reveal novel prognostic constellations enable prioritizing epigenetic therapy. Material Methods In this retrospective discovery validation study, leverage radiomic feature-driven analysis medical images supervised bioinformatic integration DNA methylation profiles. We integrate both data modalities synergistically using machine learning-based prediction cross-domain alignment. Ultimately, validate most relevant biological associations tumor tissues cell lines. Results a cohort 191 across 9 sites Austria external site South Korea, use T1-weighted contrast-enhanced magnetic resonance imaging derive score consists 20 mostly textural features. determine as strong independent predictive factor (multivariate HR=6.56), confirm its value cohort. Radiomic features align distinct, biologically meaningful ways, is predictable from selected (AUC=0.78). gene-regulatory differences between radiomically-defined groups converge on bcl6 binding activity, which posed testable treatment strategy subset patients. Conclusion The robust complementary predictor survival reflected at level PCNSL. Assessing selecting based phenotypes represents huge step forward, ability define provides concept advance modeling precision therapy for aggressive brain cancer.

Posttransplant primary CNS lymphoma.

The records and neuro-imaging studies of 8 cases of posttransplant primary CNS lymphoma (PT-PCNSL) diagnosed at Mayo Clinic Rochester between 1970 and 1998 were reviewed retrospectively. All patients received organ transplantation. Patients who had hematologic transplantation were not included in the analysis. The median and mean age of the 4 men and 4 women was 45 years (range, 34 to 50 years)...

Primary CNS Lymphoma

Primary diffuse large B-cell lymphoma (DLBCL) of the central nervous system is an aggressive malignancy that exhibits unique biological features and characteristic clinical behaviour, with overall long-term survival rates of around 20-40%. Clinical outcome has improved following the advent of chemoradiation protocols incorporating high-dose methotrexate in the mid-1980s, but disease relapse and...

Gene Expression and Angiotropism in Primary CNS Lymphoma Running Title: Gene Expression in Primary CNS Lymphoma

Primary CNS lymphoma is an aggressive form of non-Hodgkin lymphoma whose growth is restricted to the central nervous system. We used cDNA microarray analysis to compare the gene expression signature of primary CNS lymphomas with nodal large B-cell lymphomas. Here we show that while individual cases of primary CNS lymphomas may be classified as germinal center B-cell, activated B-cell, or type 3...

Treatment of primary CNS lymphoma.

OPINION STATEMENT Primary central nervous system lymphoma is a particular challenge in clinical neuro-oncology. In contrast to most other malignant brain tumors, it may be considered a curable disease at least in younger patients who can tolerate intensive treatment regimens. Yet, therapeutic progress has been limited with little measurable improvement in outcome over the last two decades, main...

Spontaneous temporary remission in primary CNS lymphoma.

We report a case of primary CNS lymphoma in which complete, though temporary, spontaneous clinical and radiologic remission occurred. This is the first such case report to our knowledge.
